Giocò stabilmente come trequartista con i neroverdi del Chieti mettendo in mostra tutte le sue ottime doti e venne subito notato dagli osservatori del Perugia. Così conobbe il grande palcoscenico della massima serie con il Perugia di Luciano Gaucci e Serse Cosmi, il trampolino di lancio verso una carriera in continua ascesa. Infatti nell'estate del 2001, Fabio Grosso firmava per il Perugia. Eravamo in Serie A. Era a scadenza di contratto ed arrivò praticamente a parametro zero. Fu poi rivenduto dal Perugia a suon di miliardi! Una delle tante plusvalenze miliardarie dell'era Gaucci-Pieroni.

Il vero successo per la carriera di Grosso è stato Serse Cosmi che, vista la sua abilità nella corsa e nel cross, da centrocampista lo ha trasformato nella sua difesa a tre, quinto di sinistra. Ed è in questo ruolo che nel 2006 segnò il rigore decisivo che consentì a tutti noi di gridare “Campioniii del Mondo!”.

Negli anni biancorossi cominciò a giocare dapprima come esterno di centrocampo nel modulo 3-5-2, e poi come terzino sinistro, disputando tre stagioni in Serie A e cominciando a imporsi a livello nazionale. Con la maglia delll'AC Perugia Calcio giocò dal 2001 al 2004 segnando 7 reti in 67 presenze e vincendo la Coppa Intertoto nel 2003.
